Isolation of compounds from Cannabis sativa and their anti-inflammatory effect

The object of this thesis is a traditional and also controversial plant Cannabis sativa. The theoretical part is focused on ubiquitous polyphenolic compounds – flavonoids, also present in the studied plant. All previously researched and confirmed effects on human body are described in detail. The experimental part of this thesis describes the isolation and identification of substances of the chloroform and hexane yields of extract obtained from this plant. Chromatographic methods were used for isolation – thin layer and column chromatography and semipreparative HPLC. Obtained substances were characterized using UV, IR spectrophotometry and NMR. Then the anti-inflammatory activity of obtained substances and also of the ethanolic extract and its yields was observed. The result of the experimental part was the isolation of one pure substance which was identified as the cannabidiolic acid. Using the THP 1 assay, we demonstrated mild anti-inflammatory effect of the non-polar yields and isolated CBDA.

The answer to what Zaježová is, can be simple. Zaježová are so called „lazy“. Zaježová is perceived by the outside world - society - primarily as an alternative community of people living in coexistence with nature, and “Zaježová” presents itself “in this way”. Personally, I think that Zaježová is defined mainly by the strong individualities of people who are able to say about themselves and present that they live in a community. Last but not least, Zaježová is a place where I spent a large part of my childhood, because my parents belonged to these strong individuals and decided to live life “alone”. My work tries to capture what Zaježová is through various media, including architecture. The output is a set of atypical tourist maps and a proposal to transform the former fire station into a gallery.
Hormones in waste waters

This bachelor thesis gives the overview of waste water contaminants of steroid hormones type. A short description of these compounds and of their physical-chemical properties is given and sources of these substances in the environment are characterized, as well as their impact on organisms The fate of these compounds and methods used for their isolation from environmental compartments methods of their final analysis are characterized.
Adjustment of the Insulation of the Bus Rear Part

Master thesis is focused on proposal of improved manufacturing process of assembly named "sandwich" produced in Iveco bus company. There is solved new fixation of assembly bench, replacement of currently used glues and material of ledge as well. In the first part there is company introduction Iveco Bus. Second part consist of construction analysis and description of present manufacturing process. Further there is introduction, solving out of thesis topics and proposal of new manufacturing process is written. At the end of thesis are attached comparison between present and proposed manufacturing process.
Multipurpose Rekreational Facility

This master thesis deals with the proposal of a multifunctional recreational facility. The complex of buildings consists of a guesthouse and stables. The guesthouse is a single-storey building with a residential attic. It also includes a restaurant with a mini-brewery and a shop selling horse riding products. The guesthouse is designed to accommodate up to 41 people. The first floor is barrier-free. The main entrance of the guesthouse and the shop faces southwest. The restaurant is accessible from the southeast. There are paved guest parking areas around the buildings. The building of stables with its accompanying facilities is a single-storey which is designed to stable 17 horses. The main entrance of the stables faces southeast and the building is also accessible from the northeast and the northwest. Both buildings have a gabled roof which consists of supporting rafters. The buildings are as designed a timber-frame construction with a supporting frame made of timber pillars which is coated with the FERMACELL boards.
Comparison of different types of magnetic carriers for DNA microisolation from foods

Micro-isolation of PCR-ready from fresh and dried legumes seeds and food products containing legumes (hummus) was tested. For optimization process magnetic microparticles PGMAox was used. Optimum weight plants and food material was 200 mg. For isolation of DNA from fresh legumes, mixture containing 500 L of CTAB extraction buffer, 1 L -mercaptoethanol and 500 L chloroform-octanol was used. For isolation of DNA from dried legumes seeds and food products volume of CTAB extraction buffer was increased to 1 000 L. To achieve higher purity of DNA some prepared homogenates was precipitate by isopropylalcohol. The optimized process of DNA isolation was used to prepare a homogenate from food products which DNA was isolated by different types of magnetic carriers. For comparison, non-porous magnetic carriers poly(glycidyl methacrylate) PGMAox, poly(2-hydroxyethyl methacrylate-co-glycidyl methacrylate) P(HEMA-co-GMA)ox covered by carboxyl groups and porous fully crosslinked microparticles poly(styrene-co-divinylbenzene) HPS-B-M-22-NH2, magnetic porous glass MPG and nanoparticles of iron oxides covered by poly(L-lysine) PLL were used. In average the highest concentration and the best purity of DNA was isolated by magnetic carriers P(HEMA-co-GMA)ox a PGMAox.
Characterization of selected yeast strains from food

This bachelor´s thesis is focused on identification of yeasts of the cheeses by PCR-RFLP method and verifying the lipolytic activity of the yeast. In the theoretical part are processed basic information about yeast, their possible positive and negative effects on the quality of cheeses, the technology of production of cheeses, lipolysis and proteolysis in the cheese and of course of PCR-RFLP (The polymerase chain reaction-restriction fragment length polymorphism). Experimental section shows the isolation of DNA, identification of DNA by PCR made by amplification 5,8S-ITS sections of DNA using primers ITS1 and ITS4. The amplified DNA was purified by ethanol and then was subjected to restriction analysis with the enzymes HaeIII, HinfI, HhaI, TaqI. Then there is listed detection of the PCR product and the restriction fragments by gel electrophoresis. Lengths of the fragments obtained after electrophoresis will be used to identify yeast species isolated from cheeses. In the second part of the thesis in the experimental part we have dealt with evidence of lipolytic activity of the yeast by test on Spirit blue agar.
Plasmid DNA interactions with lanthanoide compounds

In the theoretical part of bachelor thesis, literature was summarised on the isolation and purification of plasmid DNA, the application of lanthanides as non enzymatic nucleases and practical importance of lanthanide compounds in medicine. In the experimental part of the thesis, plasmid DNA pUC19 was isolated by the method of alkaline lysis. Isolated plasmid DNA was cleaved by neodymium trichloride at 70 °C in different intervals. Gel electrophoresis was used for evaluation of experiments.
Preparation and realization of the railway viaduct in Ostrov nad Oslavou

This Diploma thesis deals with the preparation of the realization of the railway viaduct in Ostrov nad Oslavou. Realization of major technological stages is addressed in building technological study. Technological processes of each sub-job phases are supplemented by technical regulations, the design of the main building machines and mechanisms, inspection and test plan, schedule of occupational health and safety, time and financial plan construction, construction budget, a plan for securing material resources. I also worked out the technical report of the building sites facilities including drawings needed. I also worked out design rehabilitation of the substructure of the railway viaduct, which is consist of technical regulations for remediation work and drawings.
Proposal of rehabilitation of hot water network

The bachelor thesis deals with hot water recirculation systems. It is mainly focused on the malfunctions that appear in the system. Firstly, the text elaborates general information about hot water recirculation systems, hot water distribution and its parts. The next part of the thesis describes materials used in hot water recirculation systems. Then the text deals with the malfunctions of hot water recirculation systems, design and assembly of the pipes. These chapters are followed by the case study. The case study helps to imagine the way of possible future reconstruction. The time schedule of the realization was attached to the bachelor thesis.
